捻转血矛线虫ES24抗原基因的原核表达及其免疫诊断的应用 被引量：1

摘要 将ES24基因从pUC18-ES24重组质粒中亚克隆到pET-30a原核表达载体上,构建重组表达质粒pET-30a-ES24,转化大肠杆菌,IPTG诱导表达,经SDS-PAGE电泳和Western-blot分析表明,表达蛋白相对分子质量约为30 000,可被6×His抗体和捻转血矛线虫阳性血清所识别。纯化后的重组表达蛋白作为诊断抗原具有良好的免疫反应性和特异性,可用作免疫诊断试剂。 Excretory-secretory ES24 gene from pUC18-ES24,was subcloned into prokaryotic expression vector pET-30 a and expressed in E.coli when induced by IPTG.The expressed products were tested by SDS-PAGE and Western-blot.The results showed that the expressed recombinant proteins react with 6×His antibody and H.contortus positive serum.Using the purified recombinant protein as a detectable antigen,it had a good sensitivity and specificity and could be a diagnostic reagent for H.contortus infection.
