摘要
目的木芙蓉叶有效组分(effective constituents of Hibiscus Mutabilis L.,ECHM)为从中药木芙蓉叶中提取的总黄酮,本研究旨在观察其对大鼠肾缺血再灌注损伤中肿瘤坏死因子(tumor necrosis factor-α,TNF-α)的影响。方法75只雄性Wistar大鼠随机分为对照组、木芙蓉叶有效组分(ECHM)治疗组及假手术组,每组25只。通过夹闭肾动脉60min后恢复肾脏血流灌注方法造成大鼠肾缺血再灌注损伤(renal is- chemia reperfusion injury,RIRI)模型,ECHM治疗组大鼠灌胃给药,其余两组大鼠则给予等量的生理盐水。在大鼠肾脏恢复血流灌注24h时,观察血清尿素氮(BUN)、肌酐(SCr)水平;并于大鼠肾脏恢复血流灌注的0、1、3、6h时检测血清中TNF-α含量以及肾组织中TNF-α的mRNA表达。结果再灌注24 h后,ECHM治疗组和对照组大鼠血BUN及SCr水平均较假手术组大鼠明显升高,但ECHM治疗组大鼠血BUN及SCr水平明显低于对照组(P<0.05);在肾脏恢复血流灌注的1、3h,ECHM治疗组和对照组大鼠血清中TNF-α含量均较假手术组明显升高,且ECHM治疗组大鼠血清中TNF-α含量与对照组比较差异有显著性(P<0.05);在相应的时间点,ECHM治疗组大鼠肾组织中TNF-α的mRNA表达也明显较对照组为低。结论木芙蓉叶有效组分对大鼠肾缺血再灌注损伤有一定的保护作用,可能与抑制TNF-α等炎性细胞因子的生成及活性有关。
