流式细胞仪检测胶原诱导的关节炎大鼠γδT细胞胞内TNF-α和IL-10的表达 被引量:1

摘要 目的研究牛Ⅱ型胶原诱导的关节炎模型(CIA)大鼠外周血中γδT细胞胞内TNF-α和IL-10的表达情况,探讨其在CIA中作用的分子机制。方法于Wistar大鼠背部及尾根部多点皮内注射牛Ⅱ型胶原乳剂制作CIA模型。运用流式细胞仪,通过直接荧光标记的抗TNF-α和抗IL-10抗体来检测单个细胞水平γδT细胞胞内细胞因子的表达。结果与正常大鼠相比,CIA模型大鼠外周血中γδT细胞所占淋巴细胞的百分比显著下降(P<0.01),γδT细胞胞内TNF-α(P<0.01)和IL-10(P<0.01)的表达均明显降低。结论γδT细胞在CIA中其前炎症细胞因子表达下调,可能γδT细胞在CIA的发病中不起致病作用。 Objective To detect the expressions of TNF-α and IL-10 in γδT lymphocytes in the rat model of collagen-induced arthritis (CIA) in order to discuss the possible molecular mechanism of γδT cells in CIA. Methods The experimental rat model of CIA was constructed by injection of the emulsion of bovine type Ⅱ collagen. The intracytoplasmic TNF-α and IL-10 were detected by flow cytometry and direct-labeled antibody, respectively. Results The percentage of γδT lymphocytes (P < 0.01) in the total lymphocytes in the rat model of CIA and the expressions of intracellular TNF-α ( P < 0.01) and IL-10 ( P < 0.01) of γδT lymphocytes were obviously decreased. Conclusion The γδT lymphocytes may not play a pathologic role in the mechanism of CIA for the down-regulation of the pro-inflammatory cytokines.
