背景与目的:近年研究表明,膜型基质金属蛋白酶-1(MT1-MMP)在乳腺癌的浸润和转移中有重要作用。然而,有关MT1-MMP在乳腺癌中表达的临床意义的报道极少。本研究探讨MT1-MMP在乳腺癌组织中的表达及其与乳腺癌临床病理特征的关系。方法:采用免疫组化和半定量RT-PCR的方法检测46例乳腺癌手术后标本中MT1-MMP蛋白和mRNA的表达。结果:MT1-MMP蛋白在46例乳腺癌组织中的阳性率为52.2%,其中在Ⅰ期、Ⅱ期和Ⅲ期中的阳性率分别为12.5%、54.8%和85.7%(P<0.05);在T1、T2和T3三组中的阳性率分别为11.1%、59.4%和80.0%(P<0.05);在N0、N1和N2三组中的阳性率分别为27.3%、66.7%和100.0%(P<0.05)。MT1-MMPmRNA在所有的乳腺癌标本中均有表达,其相对表达量的均数在Ⅰ、Ⅱ、Ⅲ期乳腺癌分别为0.473、0.695、0.910(P<0.05);在T1、T2和T3组中分别为0.524、0.715和0.822(P<0.05);在N0、N1和N2组中分别为0.630、0.702和0.870(P<0.05)。MT1-MMP蛋白和mRNA表达与ER、PR、c-erbB-2的状况无统计学差异(P<0.05)。结论:MT1-MMP蛋白和mRNA在乳腺癌中的表达与肿瘤分期、肿瘤大小和淋巴结转移呈正相关,有可能作为判断乳腺癌浸润转移能力的参考指标之一。
BACKGROUND &OBJECTIVE: Recent researches manifested that MT1-MM P (Membrane type-1 matrix metalloproteinase) is essential for breast cancer invas ion and metastasis,but there are few reports about the clinical value of MT1-MM P. This study was designed to investigate the clinical significance of MT1-MMP protein and mRNA expression in human breast cancers. METHODS: Forty-six human b reast cancer tissues were collected. MT1-MMP protein and mRNA expression were d etected by semi-quantitative reverse transcriptase chain reaction (RT-PCR) and immunohistochemistry assay and their correlations with clinicopathological fact ors were analyzed. RESULTS: The expression of MT1-MMP protein was positive in 5 2.2%of forty-six breast cancer tissues. The positive rates were 12.5%, 54.8% and 85.7%in stageⅠ, stage Ⅱand stage Ⅲhuman breast cancers, respectively (P < 0.05), and 11.1%,59.4%and 80.0%in the group of T1,T2 and T3 (P < 0.05), and 27.3%,66.7%and 100.0%in the group of N0,N1 and N2 (P < 0.05). MT1-MMP mRNA expression were detected in all of the carcinomas.The means of MT1-MMP mRNA exp ression relative quantities were 0.4730,0.6950 and 0.9100 in stageⅠ, stage Ⅱan d stage Ⅲhuman breast cancers, respectively (P < 0.05), and 0.5240,0.7150 and 0 .8220 in the group of T1,T2 and T3 (P < 0.05), and 0.6305,0.7022 and 0.8700 in t he group of N0,N1 and N2(P < 0.05).There were no relations between the MT1-MMP protein or mRNA expression and ER,PR or C-erbB-2(P >0.05). CONCLUSION: MT1-MM P protein and mRNA expression in human breast cancers had positive correlation w ith the stage,the tumor size and lymph node metastasis. MT1-MMP can be a predic tor for the ability of breast cancer invasion and metastasis.
