摘要
蜡质芽孢杆菌 (Bacilluscereus)BC98 I分离自沤肥浸渍液中 ,对黄瓜枯萎病等多种蔬菜土传病害有较强烈的拮抗能力。其无细胞发酵液经 5 5 %硫酸铵盐析后得到的 2 0mg ml拮抗蛋白粗提液在平皿上对黄瓜枯萎菌的抑菌圈直径达 19 3mm。该拮抗蛋白经 10 0℃处理 30min后仍能保持 97 7%的抑菌活性 ;作用活性pH范围宽 ,在pH 1 0～11 0的条件下均有活性 ;对胰蛋白酶、蛋白酶K稳定 ;对氯仿不敏感 ;对紫外线部分敏感。BC98 I拮抗蛋白粗提物对黄瓜枯萎菌孢子萌发和菌丝生长有强烈的抑制作用 ,主要表现为孢子萌发产生的芽管较对照短 ,芽管膨大形成大泡囊 ;菌丝扭曲 ,形成不规则泡囊 ,原生质浓缩 ;细胞壁溶解 ,内含物外溢。
Bacillus cereus 98-Ⅰ,isolated from compost extract made from livestock manures was used as an antagonist for biological control of plant diseases.Its crude antagonistic protein was obtained by precipitation of the cell-free culture of 98-Iwith 55% saturated (NH_4)_2SO_4.The suspension of the precipitate strongly inhibits cucumber Fusarium wilt and it is thermostable.Its active pH range is wide ,from 1.0～11.0.It tolerates trypsin, Prteinases K and chloroform.But it is sensitive to ultraviolet rays.The inhibition activity of antagonistic protein against the growth of F. oxysporum was mainly to make hyphal abnormal and sproes germination abnormally.
