A rapid, selective and sensitive LC/ESI - MSn method was developed for the quantitative determination of domoic acid in shellfish. Samples were extracted with methanol - water followed by clean-up of the extracts with strong anion exchange solid phase extraction cartridge(SPE 3 mL LC - SAX) . A reversed phase HPLC using a mobile phase of acetonitrile - water - trifluoroacatic acid( 13:86.9:0.1 % by volume) and an electrospray ionization(ESI) ion-trap mass spectromrter were used for assay. Data of domoic acid for LC/ESI - MS2 were used for quantitation. Data of domoic acid for LC/ESI - MS3 were used for confirmation. The detection limit was ca. 0.2 μg/g.
