摘要
目的 采用毛细管胶束电泳法测定血浆中异丙酚血药浓度。 方法 探讨样品预处理方法、缓冲液种类、波长、SDS浓度、pH值、工作电压、柱温对分离的影响 ,筛选后的最佳方案为 5 0mmol/L硼砂 -2 0mmol/L硼酸 -2 5mmol /LSDS -3 %甲醇 (pH =9.3 ) ,分离电压 2 0KV ,柱温 2 0℃ ,检测波长 2 72nm ,压力进样 2 0 psi× 10S。 结果 异丙酚、百里酚能得到令人满意的分离。 结论 本方法可使异丙酚血药浓度的测定时间缩短 ,方法简便、快速、灵敏、干扰少 ,是测定异丙酚血药浓度的理想方法之一。
Objective To establish a method for analysis of the determination of Propofol in plasma by micellar electrokinetic chromatography(MECC). Method By optimizing factors which affect the separation:the kinds and concentration of buffer system,pH value,voltage,temperature,the optima for separation was selected. The optimum conditions found to be 50mmol/L sodium tetraborate ,20mmol/L broic acid,25mmol /LSDS,3% methanol(pH9.3) ,working voltage at 20KV,temperature 20℃,UV detection at 272 nm,sampling: injected 10 second. Results The established method had a good seperation. Conclusion This method was simple,rapid and sensitive. It was one of the ideal methods for determination of Propofol in plasma.
