Factors Influencing on High Frequency Plantlet Regeneration of Brassica campestris ssp. pekinensis in vitro

摘要 大白菜的 4日龄子叶和 6日龄下胚轴外植体在MS +4~ 6mg·L-16 BA +0 .5mg·L-1NAA +5~ 10mg·L-1AgNO3 培养基上 3~ 4周后分化出不定芽 ,带柄子叶和子叶圆片最高再生频率均可达到 80 %~ 90 % ,下胚轴再生频率为 4 0 %~ 5 5 % ,在1/ 2MS +0 .2mg·L-1IBA培养基上 10 0 %生根 。 Cotyledons and hypocotyls of four cultivars of Chinese cabbage ( Brassica campestris ssp. pekinensis ) have been used as explants for tissue culture. Effects of ethylene inhibitor, time of seedling, and plant growth regulating substances on the frequency of shoot differentiation were investigated. High plantlet frequency regeneration system of Chinese cabbage has also been established. Factors was optimized via judicious selection of explants and manipulation of culture medium with respect to plant growth regulating substances and addition of AgNO 3 or CoCl 2 . Four day old cotyledons and six day old hypocotyls of four cultivars grown on MS medium supplemented with 4~6 mg·L -1 6 BA and 0.5 mg·L -1 NAA in the presence of 5~10 mg·L -1 AgNO 3 . Formed shoots at frequency of 80%~90% or 40%~55% after 3~4 weeks. Shoot tips rooted in half strength of MS medium containing 0.2 mg·L -1 IBA.
