摘要
目的 探讨凝血酶受体激活肽TP5 0 8和α 凝血酶对体外培养的人成纤维细胞Lu13释放转化生长因子 β(TGF β)和血管内皮生长因子 (VEGF)的影响 ,以了解其促进创面愈合的部分机制。 方法 在体外培养融合的Lu13中 ,加入不同浓度的凝血酶和TP5 0 8,置培养箱培养一定时间后 ,收集上清液离心 ,采用ELISA法测定TGF β1 、TGF β2 和VEGF的含量。 结果 α 凝血酶能显著促进Lu13释放TGF β1 ;单纯TP5 0 8使Lu13释放TGF β1 显著减少 (P <0.0 1),与小剂量凝血酶复合应用 ,对Lu13释放TGF β1 有一定协同作用。经凝血酶处理后 ,Lu13释放TGF β2 也显著增高 (P<0.0 5 )。在较大剂量 (10 0 μg ml)时 ,TP5 0 8对Lu13释放TGF β2 呈轻度抑制作用 (P >0.0 5 )。经凝血酶处理后 ,VEGF释放量显著增加 ,TP5 0 8使Lu13释放VEGF减少。 结论 凝血酶可刺激Lu13释放TGF β1 、TGF β1和VEGF,TP5 0 8抑制上述生长因子释放。TP5 0 8对TGF β1 释放的抑制作用 ,可能有助于减少瘢痕形成 。
Objective To explore the effect of thrombin peptide 508(TP508) and thrombin on the release of TGF-β and VEGF from cultured human fibroblasts(Lu13) in vitro. Methods TP508 and thrombin in different concentrations were added to the confluent cultured Lu13 and incubated for 8 to 24 hours.
