摘要
目的:探讨灯盏花素对慢性低氧大鼠肺动脉重构的影响。方法:将二级 SD 大鼠分为:正常对照组,低氧组及低氧加灯盏花素组,低氧时间为4周。采用透射电镜、免疫组化、原位杂交等方法研究灯盏花素对慢性低氧大鼠肺动脉平均压(mPAP)、右心室重量比[RV/(LV+S)]、肺细小动脉显微和超微结构、肺动脉管壁Ⅰ、Ⅲ胶原和Ⅰ、Ⅲ前胶原基因的影响。结果:(1)低氧组 mPAP、RV/(LV+S)显著高于正常对照组(P<0.01),低氧加灯盏花素组 mPAP、RV/(LV+S)显著低于低氧组(P<0.01)。(2)光镜下低氧组肺细小动脉管壁厚度占血管外径的百分比(MT%)、管壁面积占管总面积的百分比(WA%)、中膜平滑肌细胞核密度(SMC)显著高于正常对照组(P<0.01),低氧加灯盏花素组 MT%、WA%、SMC 显著低于低氧组(P<0.01);电镜检查显示低氧组肺细小动脉中膜平滑肌细胞增生,胶原纤维丰富,低氧加灯盏花素组肺动脉中膜平滑肌细胞、胶原纤维较低氧组明显为少。(3)免疫组化、原位杂交显示低氧组肺细小动脉(直径约100~200μm)Ⅰ型胶原及Ⅰ型前肢原 mRNA 平均吸光度值含量显著高于正常对照组(均 P<0.01),低氧加灯盏花素组肺细小动脉Ⅰ型胶原及工型前胶原 mRNA 平均吸光度值明显低于低氧组(均 P<0.01)。Ⅲ型胶原及Ⅲ型前胶原 mRNA 平均吸光度值各组间无明显差异(均 P>0.05)。结论:灯盏花素可能通过抑制慢性低氧肺动脉平滑肌细胞增殖和管壁胶原表达从而抑制慢性低氧肺动脉重构,降低肺动脉高压。
