摘要
目的探讨CD137分子对树突状细胞(DCs)上趋化因子分泌的调节作用。方法采用CD137-Fc融合蛋白刺激DCs,Transwell方法分析CD137-Fc诱导DCs分泌的趋化因子对不同免疫细胞的趋化作用。RT-PCR和ELISA方法分析CD137-Fc分子对DCs上不同趋化因子mRNA表达的影响。结果Transwell实验结果显示,CD137-Fc刺激的DCs培养上清对自体T细胞、单核细胞和DCs的趋化能力明显增加(P<0.05)。RT-PCR和ELISA检测结果显示,CD137-Fc刺激DCs3d后,趋化因子MIP-1α、MIP-1β、MDC和IL-8的表达水平显著增加。结论CD137可通过诱导DCs分泌MIP-1α、MIP-1β、MDC和IL-8等趋化因子促进DC募集T细胞、单核细胞和DCs等免疫细胞,进而放大DC介导的免疫反应。
Objective To study the regulation of chemokine secretion by CD137 molecule.Methods Transmi-gration assays were performed using a double-chamber transwell system. The expression of chemokines on DCs was detected by RT-PCR and ELISA analysis.Results Transwell assay revealed that CD137 could enhance the expression of chemokines in DCs.
