IL-15R α生物协同作用的体外细胞学评价

摘要 目的在细胞水平对IL-15R α的生物协同效应进行观察和评价。方法以IL-15敏感细胞株CTLL-2为靶细胞,采用MTT法检测细胞的活性及增殖情况,在摸清CTLL-2细胞增殖同IL-15之间的剂量依赖关系的基础上,分别在中、低剂量上观察经倍比稀释后的IL-15R α对IL-15体外细胞学活性的影响,其间还分别观察了IL-15R α单独作用以及IL-15R α和IL-15特异性抗体介入对CTLL-2细胞增殖的影响。结果CTLL-2细胞增殖同IL-15之间有着显著的剂量依赖关系;在50ng/mL的IL-15适中浓度下,1ng/mL的IL-15R α可以显著促进靶细胞的增殖(0.270±0.008vs.0.593±0.026,P<0.05);10ng/mL的IL-15临界浓度下,同样浓度的IL-15R α可使靶细胞产生近100倍的增殖(0.005±0.017vs.0.498±0.060,P<0.05);10ng/mL和50ng/mL的IL-15浓度下,IL-15R α发挥最大生物协同效应的浓度分别为7.81ng/mL和15.63ng/mL;单独IL-15R α没有促靶细胞的增殖效应,抗IL-15和IL-15R α的特异性抗体可有效阻断IL-15/IL-15Rα之间的协同作用。结论IL-15R α能够特异地通过IL-15发挥生物协同效应,该效应在低IL-15浓度下表现得尤为明显,IL-15/IL-15R α间适宜的配伍比例是确保该协同效应最大程度发挥的关键。 Objective To evaluate the biosynergetic effect of IL-15 receptor α subunit (IL-15R α) on cellular level. Methods IL-15 sensitive CTLL-2 cells were selected as target cells and MTT assay was adopted to determine the cell proliferation.
