摘要
目的建立胶体金标记血小板膜糖蛋白的稳定的免疫电镜技术,为开展血小板功能研究奠定基础。方法采集正常人外周静脉血,分离富血小板血浆,采用微波固定、常规固定两种方法,一抗为抗血小板GPⅢa单抗SZ21,分别选用直径10nm胶体金标记的羊抗鼠IgG(GAM-G)、直径15nm胶体金标记的葡萄球菌A蛋白(SPA-G)为探针。结果微波固定组血小板表面和开放管道系统可见到大量金颗粒的分布,而常规固定组的血小板未观察到金颗粒。结论以微波固定的方法,用大小合适的胶体金探针可特异性定位血小板表面和内部膜糖蛋白。
Objective To illustrate the fine structural localization of glycoprotein in human platelets surface, and outline our preferred methods for processing immuno-electron microscopy.Methods Platelet-rich plasma was separated from whole blood using the method of static precipitating treatment, and the samples were centrifuged to pellets, then fixation was accomplished by 1.25% glutaraldehyde using usual methods and microwave irradiation, respectively.0.05% Triton X-100 was used to permeate the platelets membrane....
