摘要 目的探讨Th1/Th2细胞因子平衡动态改变对巨噬细胞FIZZ1表达的影响。方法收集小鼠腹腔巨噬细胞,培养后将贴壁细胞分为6组,分别用不同比例的重组INFγ+重组IL-4刺激培养的巨噬细胞,RT-PCR检测巨噬细胞内FIZZ1mRNA表达,荧光免疫组化激光共聚焦显微镜检测FIZZ1蛋白表达。结果Th2型细胞因子IL-4刺激巨噬细胞,FIZZ1mRNA及其蛋白明显表达,INF-γ拮抗IL-4引起的巨噬细胞FIZZ1mRNA及其蛋白表达(INFγ+IL-4组与IL-4组比较,P<0.05),模拟Th1/Th2细胞因子平衡偏向Th2时,FIZZ1mRNA及其蛋白表达增加。结论Th2型细胞因子IL-4能刺激巨噬细胞表达FIZZ1,Th1型细胞因子INF-γ拮抗IL-4引起的巨噬细胞FIZZ1mRNA及其蛋白表达,Th1/Th2细胞因子平衡偏向Th2时,巨噬细胞FIZZ1表达增加。 Objective To explore the effect of Th1/Th2 cytokine balance switching on macrophage FIZZ1expression.Methods Collected mouse peritoneal macrophages,cultured macrophages were stimulated by different dose of recombinant INFγ+ and recombinant IL-4,adherent cells were divided into 6 groups:control groups;recombinant INFγ groups;recombinant IL-4 groups and recombinant INFγ+ recombinant IL-4 groups.
