摘要 目的 :建立人血浆中多西紫杉醇反相高效液相色谱测定法 ,为临床监测该药血药浓度提供方法学基础。 方法 :采用叔丁基甲醚 2步提取法 ,色谱条件 :岛津 SIM-PACK CL C-ODS色谱柱 ,流动相为甲醇 -四氢呋喃 -水 -氨水 (2 10∶ 7∶ 112 .5∶ 0 .0 3 ) ,紫外 2 3 0 nm检测。结果 :本法在质量浓度为 0 .0 2 95~ 5 .90μg/ ml间线性良好 ,相关系数 r=0 .99997(n=7)。日内日间 RSD均小于 5 % ,绝对回收率均大于 80 %。 结论 :本方法简便可行 。 Objective: To establish a high performance liquid chromatographic method for the determination of docetaxel in human plasma. Methods: The HPLC procedure involved a liquid liquid extraction pretreatment procedure. The sample was separated on C18 analysis column with a mobile phase of mixture of methanol tetrahydrofuran water ammonium hydroxide (210∶7∶112.5∶0.03). The UV detector wavelength was set up at 230 nm. Results: A linearity was obtained from 0.029 5 to 5.90 μg/ml of docetaxel in plasma with a good correlation ( r =0.999 97, n =7). The intra and inter day precision were less than 5%, and the absolute recoveries were more than 80%. Conclusion: The method is rapid, simple and accurate.
