谷氨酸棒杆菌S_(9114)中谷氨酸脱氢酶破碎条件的研究 被引量:2

摘要 为了研究谷氨酸棒杆菌S9114中的谷氨酸脱氢酶的特性,对细胞破碎条件对其酶活性的影响进行了研究。结果发现不同破碎条件对谷氨酸生产菌S9114细胞内谷氨酸脱氢酶的释放量不同。利用超声波对S9114细胞进行破碎时,获得GDH较好破碎效果的条件是工作时间与暂停时间比为1∶4,破壁工作时间为10min。而利用Frenchpressurecellpress进行了细胞破碎时,破碎条件为18,000Pa破碎两次。 In order to observe glutamate dehydrogenases of Corynebacterium glutamicum S_(9114),the effect of cell crush conditions on the activities of glutamate dehydrogenases(GDH) in extract is studied.The study shows that cell crush conditions affect significantly the enzymatic activities of GDH in extract.The optimized condition of cell crush is as follows:the ratio of supersonic crush and interval time is 1∶4,total supersonic crush time is 10 minutes.The cells are disrupted by passage twice through a French pressure cell press at 18,000 Pa.
