Studies on the strain selection and fermentation for producing fungal lipid

摘要 以三株被孢霉作为出发菌株 ,进行紫外线诱变 ,以乙酰水杨酸和SDS作为抑制剂 ,筛选具有抗性的突变株 .通过低温和高糖的筛选 ,再进行摇瓶试验 ,得出最适发酵条件 .通过摇瓶发酵测定各菌株的菌体干重、油脂产量 ,碘价和产不饱和脂肪酸的量 ,最终选出三个优良菌株 :A1- 16产GLA 1.5 3g/L ,ARA 1.16g/L ,DHA 0 .0 9g/L ;A2 - 6产GLA 1.11g/L ;S3- 1产GLA 1.80g/L . Using three strains of Mortierella as original strains, aspirin and SDS as inhibitors, after UV ridiation, resistant mutants were selected. By low temperature and high suger cultivation, screened out high quality strains. The optimal fermentation conditions were decided by flask experiments. Fermentation experiments determinated various strains in DCW, TL, IV and PUFA. Finally, three mutants were selected and their production: A1-16, GLA 1.53?g/L, ARA 1.16?g/L, DHA 0.09?g/L; A2-6, GLA 1.11?g/L; S3-1, GLA 1.80?g/L.
