摘要
目的:建立大鼠酒精性肾脏损害模型,观察层粘蛋白、Ⅲ型胶原在肾脏的表达。方法:以酒精灌胃法建立大鼠肾脏损伤的动物模型,应用HE染色、透射电镜等观察其病理改变;用免疫组化方法检测实验组和对照组肾组织层粘蛋白及Ⅲ型胶原的表达。结果:随着酒精刺激时间的延长,HE染色可见实验组肾脏逐渐出现肾小球系膜增殖、间质局灶性炎性细胞浸润、肾小管上皮细胞空泡变性或肿胀、管腔闭塞等改变。免疫组化染色,实验组层粘蛋白表达明显高于对照组(P<0.01);Ⅲ型胶原在两组之间无显著性差异(P>0.05)。结论:长期持续的乙醇刺激,不仅可造成肝脏损害,还可引起明显的肾脏改变且以肾小管-间质损害为重。层粘蛋白在酒精性肾损害中表达增加,而Ⅲ型胶原无改变。
Objective: Our aims were to establish a rat model of alcoholic renal injury and to observe the expressions of laminin and type Ⅲ collagen. Methods: Animal models of renal injury were established by perfusing alcohol into the rat directly. We observed the pathological changes by HE staining and transmission electron microscope. The expressions of laminin and type Ⅲ collagen in experimental group and control were measured by immunohistochemieal method. Results: By HE staining, hyperplasia of mesangium, local interstitial infiltration of inflammatory cell, vacuolar degeneration or swelling of renal tubular epithelial cell, occlusion of tubular cavity, and other pathological changes could be seen in the experimental group with the prolongation of alcohol irritation. The expression of laminin in experimental group was significantly higher than that of controls( P < 0. 01). The expression of type Ⅲ collagen had no significant difference between the two groups( P > 0. 05). Conclusion: Long-term continual perfusion of alcohol can not only damage livers but also cause renal injurys, especially to the renal tubes and interstitial area. The expression of laminin in alcoholic group increases. However, the expression of type III collagen doesn't change.
