Survivin在肝癌中的表达及其与肝癌增殖活性的相关分析

摘要 目的 研究 survivin在肝细胞癌 (HCC)中的表达及其与肝癌增殖活性的相关意义。方法 应用免疫组织细胞化学技术检测 31例肝细胞癌组织 ,8例肝硬化组织和 4例正常肝组织中survivin及增殖细胞核抗原 (PCNA)的表达水平。结果 正常肝组织和肝硬化组织中均无 survivin表达 ,31例肝细胞癌组织中 19例 (6 1.3% ) survivin表达阳性。与正常肝组织和肝硬化组织相比 ,survivin在肝癌组织中表达率显著升高 (P <0 .0 0 1)。低分化肝癌其 survivin表达阳性率显著高于高分化肝癌。此外 ,survivin表达与肝癌增殖活性显著相关 ,survivin阳性肿瘤具有较高的增殖活性 ,其 PCNA指数 30 .5 %明显高于 survivin表达阴性肿瘤 15 .8% (P <0 .0 5 )。结论 survivin在肝癌中呈现高表达 ,这种特异性表达与肝癌的低分化性和高增殖有关 ,可能涉及到肝癌的发生和进展。 Objective To study the expression of survivin and its relationship with the proliferative activity in hepatocellular carcinoma.Methods Using immunohistochemistry ABC methods, the expression of survivin and PCNA were evaluated in 31 hepatocellular carcinoma, 8 cirrhosis of liver and 4 normal livers tissues.Results No expression of survivin were detected in normal livers and cirrhosis of liver. In contrast, survivin protein was expressed in 19 of 31 patients with HCC(61.3%). When compared with normal livers and cirrhosis of liver, hepatocellular carcinoma had significantly more frequencies of survivin expression(P<0.001).The positive rates of survivin was higher in poorly-differentiated than in well-differentiated HCC. Furthermore, survivin-positive HCC had higher PCNA-LI than that of survivin-negative HCC(P<0.05).Conclusions The expression of survivin gene is increased in HCC and related to the poor differentiation, higher proliferation potential of HCC, may be involved in the onset and progression of HCC. Survivin could be a new diagnostic/therapeutic target in HCC.
