摘要
目的研究肿瘤坏死因子(TNF α)对肝星状细胞( HSC)结缔组织生长因子(connective tissuegrowth factor, CTGF)表达的影响。方法用链霉蛋白酶、胶原酶原位灌注, Nycodenz一步密度梯度离心法分离正常大鼠 HSC,并用 TNF α,转化生长因子β 1(TGF β1)处理细胞;应用逆转录聚合酶链反应(RT—PCR)技术分别观察不同培养时期的HSC中的CTGF的表达。结果 TNFα, TGFβ1均诱导HSC的CTGF表达; 10μg/L浓度的 TNFα作用 6h,没有观察到 HSC表达 CTGF;作用 24 h后,检测到有 CTGF mRNA的表达。 TGF β1在 1μg/L浓度作用 6h后,即可诱导 HSC的 CTGF mRNA表达。结论 TNF α可诱导 HSC的 CTGF mRNA表达,可能是其参与早期肝纤维化形成的作用机理之一。
Objective To investigate the effect of tumor necrosis factor α (TNF α ) on the expression of connective tissue growth factor (CTGF) in rat hepatic stellate cells (HSC) in vitro. Methods HSCs were prepared from SD rats by in situ perfusion and single-step Nycodenz gradient, and were treated with TNF α and TGF β 1.
