摘要
目的 :研究细胞周期蛋白 D1 (Cyclin D1 )和 p16蛋白在胃癌发生、发展中的作用 ,以及与胃癌生物学行为的关系。 方法 :应用免疫组化 SP法观察 Cyclin D1 和 p16蛋白在 70例胃癌组织和癌旁正常组织中的表达 ,用增殖细胞核抗原标记指数 (PCNA L I)做参数 ,分析 Cyclin D1 和 p16蛋白与胃癌预后的关系。 结果 :1Cyclin D1 蛋白阳性率 :在胃癌为 6 7.1% ,在癌旁正常组织没有表达 (P<0 .0 1) ;在高分化癌为 42 .3% ,低于低分化癌的 80 .6 % (P<0 .0 1)和未分化癌的 87.5 % (P<0 .0 5 ) ;在 PCNA L I高组为 97.1% ,明显高于 PCNA L I低组的 6 3.6 % (P <0 .0 1)。 2 p16蛋白阳性率 :在胃癌为 40 .0 % ,低于癌旁的 6 5 .7% (P<0 .0 1) ;在高分化组为 6 9.2 % ,高于低分化组的 2 7.8% (P<0 .0 1)和未分化组的 0 .0 % (P<0 .0 1) ;在 PCNA L I高组为 17.7% ,低于 PCNA L I低组的 6 1.1% (P<0 .0 1)。 3胃癌中 Cyclin D1 和 p16蛋白均阳性的 5例 ,均阴性的为 0 ,前者阳性而后者阴性的 42例 ,前者阴性而后者阳性的 2 3例 ,二者间有非常显著的关联 (P<0 .0 1)。 结论 :1在胃癌中 Cyclin D1 过表达而 p16表达缺失 ,二者呈负相关。 2 Cyclin D1 过表达及 p16缺失均与胃癌的发生和肿瘤分化程度有关 ,并且也可能与预?
Objectives:To explore the correlation of Cyclin D 1 and p16 protein expression with carcinogenesis and biologic behavior of gastric cancers. Methods:The expression of Cyclin D 1 and p16 protein in human gastric cancers and normal gastric tissues was studied immunohistochemically on 70 paraffin embeded gastric cancer tissues.PCNA LI was used to analyze the correlation between the expression of the two proteins and the prognosis of human gastric cancers. Results:①Cyclin D 1 protein positive staining was seen in 47 cancers(67.1%),whereas no Cyclin D 1 expression was found in normal tissues( P <0.01).The percentage of positive Cyclin D 1 in well differentiated carcinomas(42.3%) was lower than that in poorly differentiated carcinomas( 80.6 %)( P <0.01), and lower than that in undifferentiated group (87.5%)( P <0.05).②The expression level of p16 protein was very significantly lower in cancer tissues(40%) when compared to that in their normal counterparts(65.7%)( P <0.01).As the cancer differentiated from well to poorly the cells immunostaining p16 positive decreased very significantly in number( P <0.01).③The expression of Cyclin D 1 and p16 are adversely correlated( P <0.01).All subjects were divided into two groups by mean PCNA LI(60.9%). In the group in which PCNA LI> 60.9 %,the expression of Cyclin D 1 was higher than that in the other group( P <0.01).For p16,the expression was higher in the group in which PCNA LI<60.9( P <0.01). Conclusions:These findings suggested that the expression of Cyclin D 1 and p16 contribute to the oncogenesis,differentiation and may be related to the prognosis in human gastric cancers.And the expression of Cyclin D 1 and p16 are adversely correlated.
