摘要
[目的 ]评价 UDPE(UDG- Duplex PCR- EIA)技术在鼠疫菌检测中的效果和应用价值。 [方法 ]用 UDPE方法对鼠疫菌和其它耶尔森菌等 17株细菌及对鼠疫菌不同的菌量梯度进行检测。以鼠疫菌接种动物 ,并取动物模型的不同脏器组织同时进行分离培养和 UDPE检测比较。 [结果 ]UDPE检测系统可以特异性地对鼠疫菌进行检测 ;在梯度稀释菌液提取的模板检测中 ,灵敏度可达 0 .0 5个菌 /μl;对于接种动物的肝、脾等脏器材料阳性检出率显著高于分离培养法。 [结论 ]UDPE技术是一套特异性强、灵敏度高的检测系统 ,可有效地应用于鼠疫菌的检测和鉴定。
Objective] To evaluate the application value of UDPE method for detecting Yersinia pestis [Methods] 17 strains of Y pestis, other Yersinia and Brucella bacteria were detected by UDPE method and different concentration gradient of Y pestis To detect and compare the organ specimens (liver and spleen) of mice infected with Y pestis by UDPE and traditional cultivate method [Results] In bacteria samples, only Y pestis appeared positive result specifically For differentconcentrations levels of bacteria, the UDPE method could detect Y pestis sensitively as low as 0 05 bacteria/μl For organ specimens, UDPE detection was more sensitive than traditional cultivate method [Conclusion] UDPE method is of higher specificity and sensitivity for detecting and identifying of Y pestis
