摘要
目的 :研究肿瘤坏死因子 (TNF)对大白鼠肝的损害作用及其作用机理。方法 :随机将 2 4只雄性Sprague Dawley健康大白鼠分为两组 :Ⅰ组 (正常生理盐水对照组 ) ,Ⅱ组 (TNF处理组 )。Ⅱ组静脉注射TNF1.6× 10 6 U/kg ,处理后 0h、2h、4h、8h测血清ALT、AST、SOD、MDA ,8h后取肝组织行光镜及电镜观察。结果 :( 1)TNF组ALT、AST明显升高 (P <0 .0 5 ) ,肝组织损害 ,如肝细胞变性、坏死 ;( 2 )TNF组MDA升高、SOD降低 (P <0 .0 5 )。结论 :( 1)肿瘤坏死因子对肝脏有一定的损害作用 ;( 2 )自由基在肿瘤坏死因子介导的细胞毒性效应中起着重要作用。
Objective: To study the effect and mechanism of tumor necrosis factor inducing hepatic injury. Methods: 24 male Sprague Dawley rats were randomly devided into normal saline control group (Ⅰ)and TNF group(Ⅱ). The rats in TNF group were inject with TNF of 1.6×10 6 u/kg through vein. ALT?AST?SOD, and MAD in serum at 0, 2, 4, and 8 hours after injection were determined. The liver tissues were observed with light and electron microscope at 8 hours. Results: (1) ALT and AST in TNF group increased significantly compared with those before injection ( P <0.05).Liver tissues were found damage, including hepatocellular degeneration and necrosis. (2) MDA of TNF group increased significantly but SOD of the group decreased significantly (both P <0.05). Conclusion:(1)TNF induces obviously hepatic injury;(2)Free radicals play an important role in TNF mediated cytotoxicity.
