早期抗炎治疗对脑缺血/再灌注大鼠BDNF表达的影响 被引量:1

摘要 目的:探讨脑缺血/再灌注损伤后早期抗炎治疗对脑源性神经营养因子(Brain-derived neurotrophic factor,BDNF)表达的影响。方法:雄性SD大鼠116只随机分成5组,即假手术组(SC组)、单纯缺血/再灌注组(I/R组)、缺血/再灌注秋水仙碱干预组(Col组)、缺血/再灌注秋水仙碱和环磷酰胺联合干预组(CC组)以及缺血/再灌注维生素C干预组(VitC组)。采用线栓法制备大鼠大脑中动脉缺血/再灌注(MCAO)模型。造模成功后6h开始用药,每天给药1次,连续10d。检测BDNF在海马及梗死灶周边的表达情况。结果:I/R组大鼠海马及梗死灶周边BDNF阳性细胞较SC组和CC组明显增加(P<0.05);VitC组和Col组与I/R组比较,差异无统计学意义(P>0.05)。结论:脑缺血/再灌注诱导了海马及梗死灶周边BDNF表达增强,秋水仙碱与环磷酰胺早期联合抗炎治疗明显地抑制了这种诱导作用,而单独的VitC和秋水仙碱对BDNF表达则没有影响。 Objective:To investigate the effects of early anti-inflammatory therapy on the expression of brain-derived neurotrophic factor(BDNF) in adult rats after reperfusion in focal cerebral ischemia. Methods:116 male adult SD rats of middle cerebral artery occlusion/reperfusion(MCAO) made by intraluminal thread approach were randomly divided into 5 groups: 1) sham operation control group(SC group), 2)ischemia/reperfusion(I/R group), 3) cyclophosphamide and colchicin group(CC group), 4) colchicin group(Col group), 5)ascorbic acid group(VitC group). Medication was applied after 6 hours of operation, later once per day and last ten days.Then we detected the information of the expression of BDNF in hippocampus and peripheral focal cerebral ischemia. Results:The differences of BDNF positive cells counting in hippocampus and peripheral focal cerebral ischemia were not significant between the I/R group, Col group and VitC group(P>0.05),while compared with I/R group, the decrease of positive cells counting in the CC group was significance(P<0.05). Conclusion:Ischemia plus reperfusion can induced the expression of BDNF in hippocampus and peripheral focal cerebral ischemia;The anti-inflammatory therapy of combination of cyclophosphamide and colchicin notably inhibited the expression of BDNF. While VitC or colchicin had no effection on the expression of BDNF.
