摘要
目的:探讨建立鼠骨关节炎动物模型的方法及其可行性和可靠性。方法:出生后1周内的Wistar大鼠14只,雌雄各半,分为实验组和对照组,8只结扎其双前肢及尾部,常规饲养,分别于6、12周后随机处死1只,观察其双侧股骨头病理改变,了解效果,其余动物12周后取髋关节滑膜组织及双侧股骨头软骨为标本为实验组,6只未结扎为对照组,比较两组软骨及滑膜细胞凋亡情况。结果:12周后,无论从大体还是镜下以及细胞凋亡情况,鼠髋关节均呈典型的骨性关节炎(OA)特征。结论:Wistar大鼠是建立OA动物模型的较为理想的选择。
Objective To discuss the feasibility and dependability of the osteoarthritic animal model that get from wistar rat. Methods The anterior limb and tail of 14 rats were be ligated. Specimens of the articular cartilage and synovium obtained from 2 rat that The anterior limb and tail had been ligated for 6、12 weeks were studied by using histopathologic techniques so that the effect of the hypodesis would be observed. At 12 weeks, the samples of the articular cartilage and synovium obtained from the hip joint of 6 rats and that from another 6 rats was used to be control group. TUNEL assay was used to detect the apoptosis of the articular chondrocytes and synovial cell. Results At 12 weeks, the typical histopathologic character was seen in the articular cartilage obtained from the hip joint of rat. Conclusion It is an ideal model that to ligate the anterior limb and tail of Wistar rat to get osteoarthritic animal model.
