妊娠组织中转化生长因子及其受体与早期复发性自然流产相关性研究

摘要 目的:了解转化生长因子β1(TGF-β1)及其受体(TβR、Ⅰ、Ⅱ)在绒毛及蜕膜组织中的表达情况,初步探讨(TGF-β1)及(TβR)与早期复发性自然流产(RSA)的关系。方法:应用半定量逆转录-聚合酶链反应(RT-PCR)技术,检测38例RSA患者和35例正常早孕妇女的绒毛及蜕膜组织中TGF-β1mRNA,TβR(Ⅰ、Ⅱ型)mRNA的表达情况。结果:TGF-β1及TβR、Ⅰ、Ⅱ型在正常早孕及RSA绒毛、蜕膜组织中均有表达,TGF-β1在RSA组绒毛及蜕膜组织中表达水平为(0.18±0.02)、(0.12±0.01),对照组为(0.98±0.03)、(0.72±0.02);TβR-Ⅰ在RSA组绒毛及蜕膜组织中表达水平为(0.15±0.03)、(0.11±0.03),对照组为(0.71±0.01)、(0.73±0.01);TβR-Ⅱ在RSA组绒毛及蜕膜组织中表达水平为(0.16±0.01)、(0.09±0.01),对照组为(0.68±0.02)、(0.86±0.02);上述指标RSA组与对照组比较差异均有统计学意义(P<0.01)。结论:TGF-β1及TβR在绒毛及蜕膜组织中表达下降可能是早期复发性自然流产的重要原因之一。 Objective To understand the expressions of transforming growth factor β1 (TGF -β1) and its receptors (TβR) in villi and decidua during early pregnancy and their effects on early recurrent spontaneous abortion. Methods Expressions of TGF -β1 mRNA and its receptors were detemined in villi and deciduas by semi-quantitative reverse transcriptional-polymerase chain reaction from 38 cases of spontaneous abortion women and 35 cases of normal early pregnancy women (control group). Results TGF -β1 and its receptors had expressioned in decidua cells, villis glands and interstitial cytoplasm in all two groups. The expressions of TGF-β1 in the villi and decidua of spontaneous abortion and control group were (0.18±0.02), (0.98±0.03)and (0.12±0.01), (0.72±0.02) respectively. The intensity of TβR -Ⅰ in the villi and decidua of spontaneous abortion and control groupswere (0.15±0.03), (0.71±0.01) and (0.11±0.03), (0.73±0.01) respectively. And the expressions of Tβ1R -Ⅱin the villi and decidua of spontaneous aboetion and control groups were (0.16±0.01), (0.68±0.02) and (0.09±0.01), (0.86±0.02) respectively. The differences between the control group and abortion group were all significant (P < 0.01). Conclusion The expressions of TGF-β1 and its receptors in the early pregnancy tissues of aborted women declined. It demonstrates that the abnomal expressions of TGF-β1 and its receptors play an important role in spontaneous abortion, they are one of the causes of spontaneous abortion.
