摘要
目的探讨TGF-β3对病理性瘢痕成纤维细胞增殖-凋亡的作用。方法收集病理性瘢痕标本58例、正常瘢痕标本42例和正常皮肤30例。采用免疫组织化学法检测TGF-β3、PCNA,以及TUNEL检测标本中成纤维细胞凋亡水平。结果TGF-β3在病理性瘢痕组织中的表达低于正常瘢痕组织(P<0.05),而正常皮肤组织中TGF-β3呈阴性表达。而PCNA在病理性瘢痕成纤维细胞中表达水平与正常瘢痕及正常皮肤相比明显增高(P<0.05),正常瘢痕中成纤维细胞PCNA表达情况亦高于正常皮肤(P<0.05);另一方面,病理性瘢痕中成纤维细胞凋亡水平较正常瘢痕降低(P<0.05),而正常皮肤与正常瘢痕间无显著性差异(P>0.05);病理性瘢痕组织中成纤维细胞增殖指数增殖指数与TGF-β3呈负相关关系(P<0.05)。结论TGF-β3可能是通过调节瘢痕组织中成纤维细胞的增殖和/或凋亡水平,来实现对瘢痕形成的调节。TGF-β3表达水平的失控可能是病理性瘢痕形成的重要诱因之一。
Objective To investigate the effect of TGF-β3 to Proliferation and apoptosis of fibroblast in abnormal scar. Methods Tissue specimens were obtained from 58 abnormal scars, 42 normal scars and 30 normal skin. TGF-β3 and PCNA were stained by immunohistochemistry and TUNEL was used to detect fibroblast apoptosis. Results The positive expression of TGF-β3 was significantly lower than that of normal scar (P<0.05),but in normal skin the expression of TGF-β3 was negative Compared with normal scar and normal skin the expression of PCNA in fibroblast of abnormal scar increases significantly (P<0.05); Compared with normal skin the expression of PCNA in fibroblast of normal scar increases significantly (P<0.05). Compared with normal scar the level of fibroblast apoptosis in abnormal scar decreases significantly (P<0.05); But no significant difference between normal scar and normal skin (P>0.05). here were negative correlation with the index of proliferation and the expression of TGF-β3(P<0.05). Conclusion TGF-β3 is association closely with the occurance of abnormal scar. TGF-β3 may regulate the occurance and development of scar by means of regulating the proliferation and/or the level of apoptosis of fibroblast in scar tissue.
