1型糖尿病外周血Th1/Th2细胞因子表达水平的研究 被引量:4

摘要 目的检测1型糖尿病患者外周血CD_4^+T细胞分泌细胞因子的水平变化,探讨患者Th1/Th2细胞因子的平衡状态及其在1型糖尿病中的作用。方法对49例1型糖尿病患者和30例健康时照组外周血用刺激物刺激细胞,增加细胞内细胞因子的表达,再加入荧光标记的特异性抗细胞因子单克隆抗体,特异性抗原抗体结合,以流式细胞仪分析特异性细胞因子表达水平。结果1型糖尿病患者Th1型细胞因子干扰素-γ(IFN-γ)、白细胞介素-2(IL-2)表达水平较正常对照组升高,差异有统计学意义(P<0.01)。Th2型细胞因子白细胞介素-4(IL-4)、白细胞介素-10(IL-10)表达水平较正常时照组显著降低,差异有统计学意义(P<0.01)。结论1型糖尿病患者Th1/Th2平衡失调,Th1型反应模式处于优势状态,Th2型反应模式处于弱势状态。Th1/ Th2平衡向Th1方向漂移。 Objective To detect the levels of Thl and Th2 type cytokines of peripheral blood CD+ T lymphocytes in patients with type 1 diabetes mellitus in order to find out the role of Thl/Th2 cytokines in the pathogenesis of type 1 diabetes mellitus. Methods 49 patients with type 1 diabetes mellitus and 30 healthy children were involved in the study.The peripheral blood lym- phocytes were stimulated by stimulator in order to enhance added to be combined with its specific antigen.Cytokines were detected by flow cytometry.Results The levels of Thl type cytokines such as interferon-γ(IFN-γ),interleukin- 2(IL-2) were significant- ly higher in patients with type 1 diabetes mellitus than those in normal controls.Thelevels of Th2 type cytokines such as interleu- kin-4(IL-4),interleukin-10(IL-10) were significantly higher in patients with type 1 diabetes mellitus than those in normal con- trois.Conclusion Th1/Th2 is imbalance in type 1 diabetes mellitus.Th1 type cytokines are enhanced,and Th2 type cytokines are relatively inhibited,so Th1/Th2 shifts to Th1.It may be implicated in the pathogenesis of this disorder.
