赤芍对颈动脉球囊损伤术后兔血管内膜增生的干预作用研究 被引量:3

摘要 目的探讨高脂喂养兔颈动脉球囊损伤术后增殖的血管内膜丝裂素活化蛋白激酶磷酸酶-1(MKP-1)、还原型辅酶(NADPH)氧化酶亚单位p22phox表达、血管内皮功能及赤芍提取物的干预影响。方法喂养高脂饲料,并给予高、中、低剂量赤芍提取物(75、50、25 ml,分别含生药75、50、25 g)10周。采用Clowes方法建立兔颈总动脉球囊损伤模型。免疫组化染色测定兔颈动脉新生内膜成分,增殖细胞核抗原(PCNA)及巨噬细胞表达,血管特殊染色观察型胶原增生,图像分析系统测定新生内膜面积;张力换能器及血管环张力曲线Chart软件测定血管内皮功能;放射免疫法测定血浆血管紧张素(Ang)及醛固酮(ALD)含量;原位杂交染色及逆转录-聚合酶链反应(RT-PCR)检测增生的血管内膜中MKP-1及p22phox的mRNA表达。结果赤芍提取物可抑制型胶原增生,减少PCNA阳性细胞数,减轻新生内膜形成;降低血浆Ang水平及减弱血管对Ang的收缩反应;显著降低p22phox mRNA表达;上调高脂喂养兔颈动脉球囊损伤后MKP-1 mRNA表达,其中以高剂量组作用最明显(P<0.05或P<0.01)。结论赤芍抑制血管内膜增生作用与保护血管内皮功能有关,其机制可能涉及抗氧化应激、抗血管内皮炎症反应及阻断丝裂素活化蛋白激酶(MAPK)信号通路有关。 Objective To observe the interfering effect of Radix Paeoniae Rubra(RPR,赤芍) extract on mitogen-activated protein kinase phosphatase-1(MKP-1),reduced form of nicotinamide-adenine dinucleotide phosphate(NADPH) oxidase p22phox mRNA expressions in neo-intimal hyperplasia and its endothelial function after carotid artery balloon injury in cholesterol-fed rabbits.Methods Clowes method was applied to establish a model of balloon injury of common carotid arteries in high cholesterol-fed rabbits,and they were given ...
