摘要
目的比较两种牛颈静脉脱细胞方法。方法改良组对牛颈静脉使用0.25%聚乙二醇辛基苯基醚、2.5g/L脱氧胆酸钠、0.2g/L乙二胺四乙酸、0.1g/LRNA酶和0.2g/LDNA酶作48h脱细胞处理;常规组使用0.5g/L胰蛋白酶和0.2g/L乙二胺四乙酸处理24h,再按以上处理组程序处理24h。血管壁和瓣膜自身细胞脱除和细胞外基质改变在染色后用扫描电镜观察;血管组织强度用生物力学检测。结果改良处理组管壁和瓣膜自身细胞完全脱除而细胞外基质无明显改变,常规组仍有大量固有细胞残留,弹力纤维出现断裂;两组脱细胞血管的生物力学性能无明显变化。结论改良脱细胞处理用聚乙二醇辛基苯基醚、脱氧胆酸钠、乙二胺四乙酸、RNA酶和DNA酶组合,是一种较理想的牛颈静脉脱细胞方法。
