摘要 目的:检测海洛因对大鼠神经胶质瘤C6细胞的增殖细胞核抗原(PCNA)基因表达的影响,为探索海洛因成瘾的分子生物学机制提供依据。方法:大鼠神经胶质瘤C6细胞加入浓度为0~100mg·L-1的海洛因进行细胞培养,24h后用MTT法检测海洛因对C6细胞存活率的影响。RT-PCR法检测海洛因作用下C6细胞PCNA mRNA的表达。结果:MTT结果显示海洛因浓度为10、20和100mg·L-1时C6细胞存活率分别为90.62%、80.97%和62.73%,与对照组比较明显下降(P<0.05)。在转录物水平,浓度为20mg·L-1的海洛因作用于C6细胞48h时,PCNA基因的转录产物PCNA mRNA明显减少,与对照组比值为0.297(P<0.01)。结论:海洛因浓度为20mg·L-1时对C6细胞的增殖抑制作用接近30%,可能与海洛因成瘾形成有关。 Objective To determine the effect of heroin on the gene expression of proliferating cell nuclear antigen(PCNA) in rat C6 glioma cells,and investigate the molecular mechanism of heroin addiction.Methods The rat C6 glioma cells were treated with heroin with doses of 0-100 mg·L-1 for 24 h,MTT assay was used to detect the viability of cells,the gene expression level of PCNA was determined by RT-PCR.Results When the C6 cells were treated with heroin with doses of 10,20 and 100 mg·L-1,the cell viabilities were 90...
