摘要
目的构建一种治疗性的乙肝DNA疫苗并测定它的表达。方法采用基于表位的疫苗设计方法,用化学合成的方法制备该种分子。用适当的载体与目的分子连接后,转入大肠肝菌宿主细胞扩增。纯化后的疫苗成品转入HEK293细胞中观察它的表达。结果通过酶切反应,凝胶电泳实验及测序反应证明该质粒构建成功。转入HEK293真核细胞,能够表达。结论成功构建了一种多价抗原决定簇的治疗性乙肝DNA疫苗,且其能高效表达。
【Objective】To construct a therapeutic HBV DNA vaccine and investigate its expression in mammalian cells.【Method】 Epitope based vaccine design is adopted and chemical method is used to synthesis target genes.Target gene combined with proper vector is transformed in Ecoli cells to amplify.After the plasmid is been purified,it is transfected in HEK293 cells to observe its expression.【Result】 The plasmid is confirmed to be constructed as expectation by enzyme digestion and sequence reaction.And it can express i...
