摘要
目的探讨骨桥蛋白(Osteopontin,OPN)表达与垂体腺瘤侵袭性生长的相关性。方法分别对78例垂体腺瘤手术标本(其中侵袭性37例,非侵袭性41例)和5例来源于尸检的正常垂体组织进行检测,采用实时荧光PCR技术检测组织中OPN mRNA表达,免疫组织化学SP法检测OPN蛋白表达,结合临床资料分析OPN mRNA及其蛋白的表达与垂体腺瘤侵袭性的关系。结果侵袭性垂体腺瘤OPN mRNA表达较非侵袭性腺瘤显著增高(t=2.43,P<0.05)。OPN蛋白表达定位于细胞质,积分光密度(IOD)定量检测侵袭性腺瘤为(55.21±12.07),非侵袭性腺瘤为(48.83±13.78),两者有显著性差异(t=3.44,P<0.05)。结论OPN mRNA和蛋白在侵袭性垂体腺瘤中表达均增强,提示OPN与垂体腺瘤侵袭性生长过程有关,可能是垂体腺瘤侵袭生长的机制之一,且有望成为新的治疗靶点。
