摘要
目的探讨用凝胶成像分析系统进行DNA定量的DNA含量的检测范围。方法将已知含量的质粒DNA样本稀释成不同含量的DNA样本,进行琼脂糖凝胶电泳。利用凝胶成像系统进行观察及图像采集,并应用IMAGE J软件对DNA的电泳图谱进行灰度分析,用EXCEL软件对已知含量的DNA的灰度值与含量之间进行相关性分析,建立线性方程式。用上述线性方程式计算出样本的DNA含量。结果DNA灰度值与DNA含量之间呈正相关关系,但不成正比关系;DNA灰度值与DNA含量在一定范围内呈现良好的线性关系。通过所建立的线性关系式可以确定DNA含量的检测范围,在此范围内可较好地进行DNA的定量。结论通过建立DNA灰度值与DNA含量之间的良好线性关系,可以较好地确定凝胶成像分析系统进行DNA定量的DNA含量的检测范围。
