4-1BBL在人高转移性肺癌细胞株95D的表达及其生物学作用

摘要 目的探讨共刺激分子4-1BBL在人高转移性肺癌细胞株95D细胞的表达并在体外分析4-1BBL逆向信号对95D细胞的功能影响。方法采用免疫荧光和流式细胞术及RT-PCR检测4-1BBL分子在95D细胞中的表达,进而在体外采用可溶性4-1BB-Ig融合蛋白激发4-1BBL逆向信号,分析95D表达膜型负性免疫调节分子CD95L、PD-L1和B7-H3的变化。结果95D细胞表达4-1BBL,4-1BB激发的逆向信号可上调95D细胞表达膜型CD95L、PD-L1和B7-H3。结论体外研究结果提示共刺激分子4-1BBL表达于肿瘤细胞,4-1BBL逆向信号可能通过上调表达肿瘤细胞表面的负性免疫分子,参与肿瘤细胞的免疫逃逸机制。 Objective To analysis the expression of 4-1BBL on human highly metastatic lung cancer cell line 95D and discuss its biological effects.Methods Detecting 4-1BBL expression on 95D cell by FACS and RT-PCR,analyzing the alteration of membrane CD95L and PD-L1 on 95D cell after triggering 4-1BBL reverse signaling by 4-1BB-Ig fusion protein in vitro.Results 95D cell expressed 4-1BBL and 4-1BBL reverse signaling could regulate up membrane CD95L and PD-L1 expression.Conclusion 4-1BBL expressed on tumor cells and its...
