半边旗5F对硬皮病成纤维细胞Caspase-3活性的影响

Effects of Pteris Semipinnata 5F on Caspase-3 of Systemic Scleroderma Skin Fibroblasts

目的:探讨半边旗5F对凋亡有关的酶Caspase-3的影响,进而了解该酶诱导系统性硬皮病患者皮肤成纤维细胞凋亡的机制。方法:用光度检测法、RT-PCR法测定半边旗5F对Caspase-3的酶活性、mRNA表达量的变化。结果:在半边旗5F作用下,Caspase-3的酶活性增高,mRNA表达增多。这些变化在一定范围内随着药物浓度的增高和作用时间的延长而增强。结论:半边旗5F通过影响Caspase-3的mRNA的表达量而增高酶的活性,是诱导系统性硬皮病患者皮肤成纤维细胞的凋亡的机制之一。
