摘要
采用从山苍子油、柠檬草油等香精油分离出的柠檬醛为材料,研究MTT比色法用于筛选抗肿瘤香精油的最佳实验条件。比较不同波长选用对吸光度测定的影响;设非挥发性抗肿瘤药物阿霉素为平行实验组,以细胞形态学变化为依据,分析了不同浓度香精油药物组、对照组在96孔板上分布对MTT法测量结果的影响;此外,还探索药物储藏温度和时间对MTT法结果重复性的影响。结果表明,选择492、570、630nm三个波长对检测结果影响甚微;气态香精油物理性位移不仅使得对照组在96孔培养板上位置直接影响实验结果,而且使得低浓度组(与高浓度相邻)细胞毒性的MTT法表征值要高于实际值:挥发性香精油于低温下贮存不仅有益于保持药效,而且能确保实验重复性。
