螺旋藻β-胡萝卜素代谢通量调控规律的研究 被引量:4

摘要 结合细胞组成数据建立了螺旋藻β-胡萝卜素的代谢通量分析模型,对不同温度下培养的螺旋藻β-胡萝卜素的代谢通量进行了分析。结果显示,温度对β-胡萝卜素的代谢通量影响较为明显,33℃下通量最高,为6.51。测定了不同培养温度下β-胡萝卜素合成途径中11种相关酶的活性,并利用相关分析和主成分分析研究了酶活性对β-胡萝卜素通量的调控规律,建立了基于相关分析和主成分分析的类似与代谢控制系数的分析参数C^c_i,很好地反映了酶对通量的控制程度。结果显示,RuBP羧化酶(C^c_(RuBisCO)=0.296),葡萄糖-6-磷酸脱氢酶(C^c_(G6PD)=0.23)和丙酮酸脱氢酶(C^c_(PDHC)=0.221)对β-胡萝卜素通量具有相对较高的控制作用,说明它们对通量起着主要控制作用;α-酮戊二酸脱氢酶(C^c_(α-KGDHC)=-0.102)、磷酸甘油酸变位酶(C^c_(PGM)=-0.081)和异柠檬酸脱氢酶(C~_(ICDH)=-0.074)的系数为负值,说明它们对通量具有一定程度的负控制作用。 Metabolic flux analysis model ofβ-carotene in Spirilina was established based on data of cell composition,flux distribution of Spirilina cultured at different temperatures was analyzed according to this model.Analysis results showed that temperature has an obvious effect onβ-carotene metabolic flux,and when cultured at 33℃the flux is maximum.Activities of 11 enzymes inβ-carotene biosynthesis pathway of Spirilina cultured at a temperature gradient were measured,and then correlation and components analysis we...
