维生素C对硼替佐米诱导多发性骨髓瘤细胞株KM-3凋亡作用的影响 被引量:2

摘要 目的:为了研究维生素C对硼替佐米诱导多发性骨髓瘤细胞株KM-3细胞凋亡效应的影响。方法:采用细胞计数法检测硼替佐米15nmol/L+10、20、40、80μmol/L维生素C处理KM-3细胞生长抑制率,用流式细胞术分析Annexin-V/PI、线粒体跨膜电位(Δψm)和氧自由基(ROS),用分光光度法检测Caspase3活性。结果:硼替佐米对KM-3细胞增殖抑制作用降低,24 hAnnexin-V阳性细胞比例下降,△ψm降低被阻止,18h活性氧水平下降,16hCaspase-3活性下降。结论:维生素C对硼替佐米诱导KM-3细胞凋亡效应具有抑制作用。 Objective: To investigate the effect of Vitamin C on multiple myeloma cell line KM -3 apoptosis induced by Bortezomib. Methods: Cell viability inhibition ratio was estimated by counting method. Annexin -V,mitochondrial transmembrane potential(Δψm) and reactive oxygen species(ROS) labeled by DCFHDA were examined by flow cytometry, and the activity of Caspase-3 was detected by spectrophotometry.
