摘要
用合成的线性多肽探讨Graves病(Graves disease,GD)小鼠模型的免疫状态。用表达促甲状腺素受体(TSHR)A亚单位的腺病毒(A-TSHR-Ad)免疫BALB/c雌性小鼠获得GD小鼠模型;用合成的TSHR A亚单位的线性多肽PA,刺激小鼠脾脏细胞,检测记忆性淋巴细胞反应中生成的细胞因子。所有接受免疫的小鼠均出现TRAb增高,66.7%的小鼠出现甲亢;接受免疫的小鼠脾脏细胞在接受多肽刺激后,IFN-γ生成增加(PPA-2=0.01),IL-4的生成则未受影响。表明T细胞可以识别TSHR A亚单位的线性多肽PA,多肽PA可以促进GD小鼠Th1免疫应答的发生。
To investigate the immune responses in Graves disease(GD) mouse model with synthetic linear peptides,the female BALB/c mice were immunized with adenovirus expressing the A subunit of TSH receptor(TSHR)(A-TSHR-Ad),thus producing the GD mouse model.The splenocytes of this mouse model were stimulated with synthetic linear peptides,and the cytokines produced in the culture supernatants of the stimulated memory lymphocytes were assayed.It was found that all the immunized mice showed an elevated level of TRAb wi...
