千金黄连丸加味方对胰岛素抵抗小鼠胰腺的影响 被引量:8

摘要 目的探讨中药复方千金黄连丸加味方对谷氨酸钠(monosodium L-glutamate,MSG)诱发的胰岛素抵抗(insulin resistance,IR)模型小鼠胰腺的影响,以及其作用的分子机制。方法以中药复方千金黄连丸加味方干预MSG IR小鼠模型,与罗格列酮(rosiglitazone,Ros)作对照,测定小鼠的空腹血糖、血清胰岛素水平及胰岛素敏感指数,观察胰腺形态学变化,并以逆转录-聚合酶链反应(RT-PCR)方法测定胰腺组织中胰岛素受体、胰岛素受体底物(insulin receptor substrate,IRS1/2)及葡萄糖运载体(glucose trans-porter,GLUT2)mRNA表达。结果中药组空腹血糖较模型组明显下降(P<0.01),胰岛素敏感指数较模型组升高(P<0.05),与罗格列酮组比较差异无统计学意义。中药组胰岛的增生及肥大减轻,与模型组比较差异有统计学意义(P<0.01),与罗格列酮组比较差异无统计学意义。中药组小鼠胰腺组织中GLUT2mR-NA的表达量较模型组明显增加(P<0.01),罗格列酮组无明显改变。结论中药千金黄连丸加味方能减轻胰岛素抵抗,改善胰腺病理变化,其作用与增加胰腺组织GLUT2mRNA的表达,从而改善了胰腺的IR有关。 Objective To investigate the influence of modified Qianjin Huanglian Pill(QJHL),a Chinese herbal compound,on pancreas in mice with monosodium L-glutamate(MSG)induced insulin resistance(IR)and its molecular mechanism.MethodsControlled by rosiglitazone(Ros),the MSG indiced IR mice were treated with QJHL for 28 days.The laboratory indices were examined including fasting serum glucose(FSG),fasting serum insulin(FSI),insulin sensitivity index(ISI),and morphological changes of pancreas,and levels of insulin recep...
