摘要
以菠萝蜜的叶片和花序为材料,采用改良的十六烷基三甲基溴化胺(CTAB)法(在原来的提取方法基础上,使用乙酸钾和无水乙醇预先沉淀果胶),成功提取菠萝蜜叶和花序的总RNA,平均产率分别为74.7μg/gFW和31.6μg/gFW。经紫外分光光度计检测、1%琼脂糖凝胶电泳和RT-PCR扩增分析结果表明,使用该方法所提取的RNA质量高,能够满足相应的分子操作。
Using modified apple fruit RNA extraction method based on CTAB by adding a step of using KAc and absolute ethanol to precipitate pectin, high-quality total RNA was extracted from the leaf and the inflorescence of jackfruit (Artocarpus heterophyllus Lam.). The yields of total RNA were 74.7 μg/g FW for the leaf and 31.6 μg/g FW for the inflorescence. Spectrophotometer, agarose gel electrophoresis and RT-PCR showed that the total RNA isolated by this method was of high quality and could be used for RT-PCR.
