摘要
介绍目前已广泛应用于建兰花叶病毒(Cymbidium mosaic virus,CymMV)和齿兰环斑病毒(Odontoglossumringspot virus,ORSV)检测的电镜技术、血清学方法和分子生物学方法,以及以这3种方法为基础建立起的酶联免疫吸附反应法(enzyme linked immunosorbent assay,ELISA)、免疫胶体金技术、免疫毛细区带电泳法(Immuno-capillaryzone electrophoresis,I-CZE)、反转录聚合酶链式反应法(Reverse transcription-polymerase chain reaction RT-PCR)、免疫捕捉PCR技术(IC-RT-PCR)、TaqMan实时RT-PCR技术。同时介绍石英晶体微天平生物传感器(Quartz Crystal Microbalance biosensor,QCM biosensor)应用检测技术、液相色谱-质谱分析和激光解吸-离子质谱分析法(LC-and MALDI-mass spectrometry)2种新检测技术。并简述了各种检测方法的优缺点及应用范围。
This paper reviews the microscopical,serological and molecular means for detecting of both CymMV and ORSV.The technologies of ELISA,Immunogold-label assay,I-CZE,RT-PCR,IC-RT-PCR and TaqMan Real-Time RT-PCR established based on the microscopical,serologival and molecular means,and two other new detection methods: QCM immunosensors and LC-and MALDI-mass spectrometry are summarized.The advantage and disadvantage in application of these detection methods are discussed.
