摘要
目的:观察苦参碱诱导胶质瘤模型大鼠胶质瘤细胞凋亡的作用并探讨其机制。方法:采用脑立体定位仪大鼠脑内注射C6细胞法建立胶质瘤模型,通过实验大鼠的生存状态、取材标本的大体所见、MRI检测及组织切片HE染色观察苦参碱对胶质瘤大鼠模型生存质量、胶质瘤体积及胶质瘤细胞凋亡的影响;采用免疫组织化学(IHC)及Western-blot方法检测苦参碱对胶质瘤大鼠胶质瘤细胞BID表达的影响。结果:实验大鼠的生存状态、取材标本的大体所见、MRI检测及组织切片HE染色结果显示苦参碱可显著提高胶质瘤模型大鼠的生存质量、抑制胶质瘤细胞增殖并诱导其凋亡;IHC及Western-blot结果显示苦参碱可诱导胶质瘤模型大鼠胶质瘤细胞BID表达的上调。结论:苦参碱具有抑制胶质瘤模型大鼠胶质瘤细胞增殖并诱导其凋亡作用,其作用可能是通过凋亡的线粒体途径上调BID的表达来实现的。
Objective:To observe the effect of matrine on rats’ glioma-cell apoptosis and discuss its mechanism.Methods: Glioma models were made by injecting glioma-cell suspension in rats’ brains located by stereotaxic apparatus. The rats’ life quality, glioma volume and apoptosis of glioma-cell were observed respectively by survival state, gross changes of tumor, MRI and HE staining. BID expression in glioma-cell of each group was detected by Western-blot and immunohistochemistry methods.Results: The results showed that matrine could significantly improve the life quality of rats with glioma, inhibit the proliferation of glioma-cell and induce their apoptosis in vivo by the observation of survival state, gross changes of tumor and examination of MRI and HE staining. The results of Western-blot and immunohistochemistry test showed that matrine could up-regulating BID expression in glioma-cell.Conclusion: Matrine can inhibit the proliferation of glioma-cells and induce their apoptosis in vivo, which maybe complete by instrinsic mitochondrial pathway of apoptosis, especially up-regulating the expression of BID.
