摘要
目的:探讨化瘀濡肺汤对博来霉素A5所致大鼠肺纤维化的干预作用及其机理。方法:雄性Wistar大鼠60只,随机均分为正常对照组、模型组、中药高剂量治疗组、中药低剂量治疗组。大鼠气管内一次性注入博来霉素A5制成肺纤维化模型。模型制备后第2天开始,分别给予相应处理,4组动物分别于第7、14、28天各处死5只,提取肺组织,应用免疫组化法检测肺组织内转化生长因子β1(TGF-β1)及Ⅰ、Ⅲ型胶原的表达。结果:与模型组比较,中药低剂量、高剂量治疗组肺泡炎、肺纤维化的程度明显降低(P<0.01),肺组织内Ⅰ、Ⅲ型胶原及TGF-β1的表达也明显减少(P<0.01)。结论:化瘀濡肺汤可有效减轻大鼠肺纤维化,抑制转化生长因子β1及Ⅰ、Ⅲ型胶原在肺组织中的表达是其可能的作用机制之一。
Objective:To discuss the intervention effect and the machanism of huayu rufei decoction on rats’ pulmonary fibrosis induced by bleomycin A5.Methods: 60 male Wistar rats were divided into normal,model,high and low dose decoction groups at random,each group included 15 rats.Rats’ pulmonary fibrosis models were made by single intracheal instillation of bleomycin(5mg/kg), each group rats were given corresponding drugs respectively,5 rats were killed in each group after modeling 7,14 and 28 days,the expressions of TGF-β1,collagen Ⅰ and Ⅲ in lungs of each group rats were determined by immunohistochemistry method.Results: To compare with the model group,the severity of rats’ lung inflammation and fibrosis were lightened in the two Chinese herbs groups,the expressions of TGF-β1,collagen Ⅰ and Ⅲ in lungs were reduced more obviuosly.Conclision: Huayu rufei decoction can relieve pulmonary fibrosis,inhibiting the expressions of TGF-β1,collagen Ⅰ and Ⅲ in lungs maybe one of its mechanism.
