氧化苦参碱联合氟尿嘧啶对结肠癌LoVo细胞的影响

Effect of Oxymatrine Combined with Fluorouracil on Human Colon Carcinoma Cell Line LoVo

目的:观察氧化苦参碱(oxymatrine,OM)与氟尿嘧啶(fluorouracil,5-FU)联合应用对结肠癌LoVo细胞的影响及其作用机制。方法:MTT法测定单用OM或5-FU及两者联用对LoVo细胞的毒性作用的影响,并计算合并指数;流式细胞仪检测OM与5-FU联用对LoVo细胞凋亡的影响。结果:5-FU联用IC_(10)浓度的OM时对LoVo细胞的抑制作用比单用5-FU时增效2.67倍,OM联用IC10浓度的5-FU时比单用OM时增效2.01倍,两药的合并指数为0.87;5-FU和OM联用后,细胞凋亡率比单用时分别增加了2.08及1.82倍。结论:氧化苦参碱与氟尿嘧啶体外联用有协同抗结肠癌LoVo细胞的作用。
