Parthenolide对急性单核细胞白血病细胞株THP-1增殖与凋亡影响的观察

摘要 目的:探讨Parthenolide(PN)对急性单核细胞白血病细胞株THP-1的细胞增殖作用及其可能的作用机制。方法:CCK-8法检测不同浓度处理的THP-1细胞的半数抑制浓度(IC50);以6μmol/L的PN作用于THP-1细胞,设正常细胞对照及DMSO溶剂对照,以RT-PCR检测各组细胞核因子κB(NF-κB)p65因子和阻遏物IκB的mRNA表达,以蛋白质印迹法检测p65和IκB全蛋白的表达及p65核蛋白的表达。结果:PN对THP-1细胞有增殖抑制作用,PN作用12h的IC50为8~10μmol/L,24h的IC50为6~8μmol/L;1~10μmol/LPN的抑制率随浓度的增加而加大,存在剂量-效应关系,最大抑制率12h为(69.56±2.52)%,24h为(68.20±2.04)%。6μmol/LPN作用于细胞24h,与两对照组相比,p65mRNA及总蛋白的表达差异无统计学意义,但核蛋白表达明显下降,P<0.05;IκBmRNA及总蛋白的表达均有明显上调,P<0.05。结论:PN可通过上调细胞IκB表达,阻滞p65由细胞质向细胞核转运,从而下调NF-κB通路活性,而抑制THP-1细胞的增殖。 OBJECTIVE: To study the effects of parthenolide(PN) on proliferation and apoptosis of THP-1cells and investigate the related mechanisms.METHODS: The growth inhibiting rates were assayed by the CCK-8 method,the expression of NF-κB p65 and IκB mRNA were examined by real time-PCR,and the protein level of NF-κB p65 and IκB were detected by Western blotting in the cells of defferent groups.RESULTS: PN inhibited the cell proliferation of THP-1 cells,and IC50 of THP-1 cells affected by PN at 12 h were 8-10 μmol/L ...
