摘要
通过台盼蓝拒染法,以细胞存活率为指标用扫描电镜观察超声激活原卟啉Ⅸ(ProtoporphyrinⅨ,PPⅨ)对艾氏腹水肿瘤(Ehrlich ascites tumor,EAT)细胞的杀伤作用;采用硫代巴比妥酸法检测超声激活PPⅨ对EAT细胞膜脂质过氧化水平的影响,研究超声激活Ⅸ对EAT细胞的杀伤效应.结果显示,超声结合PPⅨ杀伤EAT细胞与超声强度、处理时间及PPⅨ药物浓度呈正相关,超声结合PPⅨ对EAT细胞的损伤显著高于单纯超声组,而单纯PPⅨ表现出无明显的细胞毒作用;超声激活PPⅨ后膜的脂质过氧化水平显著增加.实验表明超声激活PPⅨ对EAT细胞具有明显的协同杀伤效应,推测可能是超声激活PPⅨ增加了膜脂质过氧化量,从而使细胞受损,并且质膜可能是超声激活PPⅨ杀伤EAT细胞的主要靶点之一.
Typan blue exclusion method and the scanning electron microscope were used to analyze the viability of Ehrlich ascites tumor(EAT) cells in presence of protoporphyrin Ⅸ disodium salt(PP Ⅸ) ultrasonically activated.The level of lipid peroxidation was detected by using thiobarbituric acid reaction method.The cytotoxic effect of PP Ⅸ induced by ultrasound on isolated EAT cells were investigated.The results indicated that no significant effect could be checked out in the cells treated by PP Ⅸ.The cells treated b...
