蛛网膜下腔移植APA-NIH3T3/rPENK对大鼠神经痛的镇痛效应 被引量:4

摘要 目的:探讨海藻酸钠-聚赖氨酸-海藻酸钠(APA)微囊化转鼠前脑啡肽原基因NIH3T3细胞(APA-NIH3T3/rPENK)移植对大鼠神经痛的镇痛作用。方法:50只SD大鼠随机分为5组:CCI(坐骨神经慢性压迫)组、假手术组、APA空囊组、NIH3T3/rPENK组和APA-NIH3T3/rPENK组。测定CCI组和假手术组手术前后,APA空囊组、NIH3T3/rPENK组和APA-NIH3T3/rPENK组移植前后CCI术侧热痛阈,观察腹腔注射纳洛酮对细胞镇痛效应的影响,同时用放射免疫法检测脑脊液灌流液中亮氨酸脑啡肽(L-EK)含量。结果:CCI术后7~33d术侧后爪热痛阈明显低于非术侧后爪(P<0.05)。APA空囊组移植前后热痛阈未见明显变化;移植后NIH3T3/rPENK组和APA-NIH3T3/rPENK组热痛阈都明显高于APA空囊组(P<0.05)。移植15d后NIH3T3/rPENK组热痛阈显著低于APA-NIH3T3/rPENK组(P<0.05)。移植21d后APA空囊组脑脊液灌流液中L-EK含量显著低于NIH3T3/rPENK组和APA-NIH3T3/rPENK组(P<0.05);NIH3T3/rPENK组脑脊液灌流液中L-EK含量显著低于APA-NIH3T3/rPENK组(P<0.05)。结论:NIH3T3/rPENK或APA-NIH3T3/rPENK植入大鼠的蛛网膜下腔可以明显减轻神经痛大鼠的热痛敏感行为。APA-NIH3T3/rPENK效果更明显,提示APA微囊具有良好的免疫隔离作用。 Aim:To study the analgesic effect of intrathecal APA microencapsulated NIH3T3/rPENK on neuropathic pain in rats.Methods:50 SD rats were randomly divided into five groups:CCI group,sham-operated group,APA group,NIH3T3/rPENK and APA-NIH3T3/rPENK groups.The thermal threshold of the operated hindpaws was examinated before and after transplantation in APA,NIH3T3/rPENK and APA-NIN3TE/rPENK groups.The analgesisc effect of neuropathic pain was observed after infecting of naloxone in rats,The concentrations of enkep...
