经冠脉自体骨髓间充质干细胞移植对心肌梗死兔血管新生和心脏功能的影响 被引量:2

摘要 目的探讨经冠脉注射法移植自体骨髓间充质干细胞(BM-MSCs)对兔急性心肌梗死(AMI)心脏的血管新生和心脏功能的影响。方法取日本大耳白兔45只,随机分为BM-MSCs移植组、对照组和假手术组,各15只。BM-MSCs移植组抽取骨髓液分离、纯化BM-MSCs并体外扩增,结扎兔冠脉左前降支(LAD),建立AMI模型。再灌注后,BM-MSCs移植组经冠脉注射自体BM-MSCs悬液1ml(5×106个)。对照组以相同的方法建立AMI模型,注入等量的无血清培养液。假手术组除不结扎LAD外,其余操作同对照组。术后24h和4周,以超声心动图仪测量各组左室射血分数(LVEF)和左室短轴缩短率(FS)。4周后,通过抗5-溴脱氧尿核苷(BrDU)抗体免疫组化染色评价移植细胞的存活情况,并测定各组血管内皮生长因子(VEGF)蛋白的表达及毛细血管密度的改变情况。结果术后4周,假手术组的LVEF和FS分别为(62.1±2.5)%、(32.2±1.7)%,对照组分别为(53.2±2.3)%、(23.8±0.8)%,BM-MSMs移植组分别为(62.4±2.6)%、(30.8±1.9)%,对照组的LVEF和FS与BM-MSCs移植组比较差异有统计学意义(P<0.01),与假手术组比较差异亦有统计学意义(P<0.01),而BM-MSCs移植组的LVEF和FS与假手术组比较差异无统计学意义(P>0.05)。BM-MSCs移植组在梗死区可见抗BrDU抗体染色阳性细胞。假手术组的新生血管数目为(2.0±1.2)个,对照组为(4.1±1.8)个,BM-MSCs移植组为(12.5±1.7)个;病理图像测量系统测定平均光密度值(400×),假手术组VEGF蛋白表达为(0.07±0.01),对照组为(0.11±0.01),BM-MSCs移植组为(0.15±0.03);BM-MSCs移植组的新生血管数目、VEGF蛋白与假手术组比较,差异有统计学意义(P<0.01),BM-MSCs移植组与对照组比较,差异有统计学意义(P<0.01)。结论经冠脉注射法移植自体BM-MSCs可在AMI区存活,增加VEGF蛋白表达,诱导血管新生,改善心脏功能。 Objective To observe the influence of autologous bone marrow mesenchymal stem cells(BM-MSCs)transplantation by intracoronary infusion on angiogenesis and cardiac function in myocardial infarction rabbits.Methods Forty-five white Japanese big-ear rabbits were divided randomly into 3 groups,BM-MSCs,control and sham operation,15 in each.In BM-MSCs group,bone marrow was extracted to establish AMI models by separating,purifying and in vitro amplifying BM-MSCs,and ligating left anterior descending(LAD)of coronary...
